[JCRB4190 AIK1: passage procedure]

Reagents

●Culture medium: Keratinocyte SFM (1x) kit (Gibco Cat.# 17005042)
●PBS(-)
●0.25%Trypsin-0.02%EDTA (in PBS(-))
●1% Trypsin inhibitor in PBS(-)*
* Dissolve trypsin inhibitor (Gibco cat.No.17075029, 7000 BAEE unit/mg protein)(powder) in PBS(-) to 1％, filter sterilize, dispense into small alquots, and store frozen. Dilute to 0.25% in PBS(-) before use.

Passaging (for 60-mm dish)

1.	Wash the culture with PBS(-) once.
2.	Add 2 mL 0.25%Trypsin-0.02%EDTA to dish, and incubate at 37 C for 3 – 5 min. (The cells will become round shape. Avoid the long treatment until the cells completely detach from dish, but the treatment should be stopped just after the cells start to detach.)
3.	Terminate the trypsin activity by adding 2 mL of 0.25% trypsin inhibitor solution, and collect the cell suspension to centrifugation tube.
4.	To collect the cells remaining in dish, wash the dish with 5 mL culture medium and transfer the medium to the centrifugation tube(3)
5.	Centrifuge at about 200 xg for 4 - 5min.
6.	Discard the supernatant and gently tap centrifugation tube to loosen the cell pellet. Then resuspend the cells to fresh culture medium and seed to new dishes at split ration 1/3 – 1/4. The suitable seeding cell density will be 1 – 2 x 10^4 cells/cm2. Pssage interval will be 4 – 5 days.

Note
The keratinocytes are weak to trypsin treatment, and too long treatment often causeｓ the growth problem. The treatment time should not exceed 5 min.

用意するもの

●培地: Keratinocyte SFM (1x) kit (Gibco Cat# 17005042)
●PBS(-)
●0.25%Trypsin-0.02%EDTA (in PBS(-))
●1% Trypsin inhibitor in PBS(-)※
※ Trypsin inhibitor (Gibco cat.No.17075029, 7000 BAEE unit/mg protein)(粉末)をPBS(-)で1％になるよう溶解し、ろ過滅菌する。分注して凍結保存し、使用時にPBS(-)で４倍に希釈(0.25%)する。

継代 (60-mm dishの場合)

1.	PBSで一回洗浄する。
2.	0.25%Trypsin-0.02%EDTAを2mL添加し、３７℃インキュベータの中に3～5min置く。(細胞が丸みをおびてくる。この際、細胞が完全に浮遊するまでは処理せず、浮き上がる直前までの処理が良い。)
3.	0.25% Trypsin inhibitorを2mL添加してトリプシンの活性を止め、細胞浮遊液を遠心管に回収する。
4.	Dishに残っている細胞をさらに回収するため、培地5mLでdishを洗い、3の遠心管に加える。
5.	200 xg程度で4-5min遠心する。
6.	上清をすてペレットを軽くタッピングしてほぐした後、1/3～1/4splitになるよう新しいdishに播種する。(播種密度：1～2 x 10^4/cm2, 継代頻度: 約4～５日ごと。)

注意点
ケラチノサイトはトリプシン処理に弱く、長時間作用させると増殖不良を招きやすい。トリプシンの作用時間は5 minを超えないよう注意する。
