[On the cultivation of JCRB9119 Ramos(RA-1)]

Ramos(RA-1) seems wead for freeze-thawing, and many dead cells will be observed after thawing.

The survival of cells will be dependent on the cell density. Therefore please take the following procedure at the re-cultivation of cells from frozen vial.

1) Suspend the all vial contents to 10 mL culture medium containing FBS after thawing.
2) To remove DMSO, centrifuge the cell suspension at 1200 rpm for 5 min, and remove the supernatant.
3) Resuspend the cells to 5 mL of fresh culture medium with FBS.
4) Plate the all cell suspension to one well of 6 well plate.
5) Ramos cells will rapidly grow when they start the proliferation. To avoid the overgrowth, please prepare the additional culture by dilution (take 1 mL of cell suspension and suspend to 4 mL fresh merium; 1/5 dilution). Original culture should also be continued to cultivate.

Ramos (RA-1)は凍結融解に弱い傾向があるとみられ、解凍後多数の死細胞が観察されます。

解凍後の生存性は細胞密度に依存しますので、次の手順をとってください。

1) バイアルを解凍後、10 mlの血清入り培地にサスペンド
2) 1200 rpm, 5分遠心し、上清を除く(DMSOを除くため)
3) 細胞を5 mLの血清入り培地に再浮遊
4) 6 ウェルプレートの1ウェルにて培養を開始
5) 増殖が開始されると増殖速度は速いため、解凍翌日、細胞の状態をみて、1 mLをとり、1/5希釈した培養も用意してください。
